Yes and no, the film shares the same source material, the manga 'Litchi Hikari Club'. The film follows the manga storyline closely, while the animated series is a parody of the manga, having a lighther tone and does not follow the storyline. Note that this film is not a comedy. It is a psychological horror like it's source material.
They are all of middle school age. Most likely 13-14 years old, as Zera and the others are all in the same grade and Zera comments that he "will be turning 14 tomorrow". The actors who portray them are all in their 20s-30s.
Zera has an imaginary friend in the film who replaces The Fortune Teller. Tamiya's little sister Tamako does not appear in the film, her rape is replaced by Kanada's death. Kanada is killed when Tamiya is forced to choose between his own and Dafu's death, opposed to being executed when Jaibo frames him for breaking the chess piece. Niko does not return for the final battle, it is implied he died shortly after he was rescued. Zera is killed by Lychee, not Niko. Zera's death is changed due to this, he is not impaled by a urinal like in the manga, but by Lychee's fist. Kanon does not survive, like she does in the manga.
this does contain spoilers to bonus scenes !!! the movie does contain some scenes from the prequel novel so i'll be mentioning that as well in the manga, the weapon that supposedly kills dafu (SPOILER dafu did survive but was in a coma for 10 ? years) is actually supposed to be a slingshot, not sure why they replaced it but it was a slingshot, something tamiya was very skilled at using. kanon in the movie is much more resentful towards the group compared to any other adaptation, constantly trying to escape and fight back. in the manga (and other versions) she doesn't seem to mind being there, she believes their actions are wrong but doesn't fight as much. they skip out on a lot of side character things, unfortunately they don't talk much about niko in the movie and skipped out on a lot of his scenes (for example, him and tamiyas friendship, they were good friends and tamiya cares deeply for niko but they don't expand on that). There are also different versions of dafu being discovered with kanon, in some versions its zera who finds him, in others its litchi

Zera's imaginary friend replaces The Fortune Teller seen in the manga. It, unlike The Fortune Teller, tells Zera to become godlike, opposed to Zera taking that understanding from what his fortune prophesied. It is seen at the beginning talking to ten year old Zera telling him that Zera is not like other children and will eventually rule humanity after it helps Zera kill anyone who stands in his way.
Tamiya, Kanada, and Dafu created the Light Club when they were in elementary school. Tamiya proclaimed himself the leader as he named it after finding that by taking two kanji from each of their names spelled light (hikari). Later on, Zera joins, when Tamiya promises to teach him to play chess and be his friend. Zera and the others decide to create a large robot of their own after Zera creates a toy prototype for a class project. Zera takes the project to heart believing that with a robot with human capabilities he can eventually dominate the world. The club is aware of this, but believes it to be a game, as they are young children. Along with plotting world domination, Zera begins to plan out the genocide of adults, believing that growing up ruins a person, making them 'ugly'. As the project progresses, Zera proclaims himself emperor of The Light Club and creates his own set of commandments gaining complete control of the members.
Zera came from a home where his father left the family and his mother was depressed. He grew up hearing his mother refer to men as 'sloppy' and 'disgusting' as well as watching her sink farther into depressed mental state. Observing this and having little interaction with other adults or children, Zera believes that was what adult life was like and he begins to detest adults and the concept of adulthood. He, by becoming emperor of The Light Club, along with the members of the club who vowed to never grow up, would create a new world where they never they never grow up, and in turn, stay beautiful. To obtain this goal, the club sets out to execute adults for the sin of growing up and becoming ugly.
Zera believes that The Light Club needs balance as all it's members are all male. He orders Lychee to kidnap a beautiful girl so that she may balance him as empress.
