As Ishaan, a music producer who has commitment issues with the women in his life, feels a connection with Gauri, his next-door neighbor, he begins to learn the importance of relationships.

Ishaan (John Abraham) a charismatic, good-looking music producer from Mumbai, lives a sheltered existence as the apple of his mother Nisha's (Zarina Wahab) eye, always protected by his elder sister Shivani (Mini Mathur). Nisha has taught Ishaan since he was a child, that he is the best and that he can do no wrong.

He lives with his girlfriend Anushka (Chitrangada Singh) and takes her for granted. He stays with Anushka and even refuses to share the groceries bills. He tells Anushka that he is so good looking that any girl would be happy to do what she is doing for him (she cooks him breakfast daily).

Beena (Raima Sen) is Ishaan's boss at work. Beena keeps rejecting all of Ishaan's finds. She says that they are nothing more than bimbos that he wants to have sex with at the company's expense. Beena says that she was hired laterally, and made Ishaan's boss, even though Ishaan was with the company for many years, so that he could be put in his place. Ishaan introduces an emerging talent named Sukanya to Beena, but she insults Sukanya (by making her sing "Happy Birthday" song as an audition for her 5-year-old dog's party) and kicks her out.

Anushka is good friends with Shivani. Shivani tells Anushka to dump Ishaan, but she is sure that she loves Ishaan dearly. Shivani insists that Ishaan is not ready for the responsibility of marriage. Shivani is married to Adil (Mukul Chadda) and has a son named Rishi (Krish Chatterji). Ishaan throws a house party for his office colleagues without telling Anushka and then she was left to clean up the place, including taking care of guests who end up vomiting in her house. Anushka wants Ishaan to take her to meet his parents in Pune, but Ishaan completely forgets and keeps Anushka waiting. That night Anushka kicks him out of her house due to his irresponsibility about their relationship. Anushka realizes that the relationship is a joke to Ishaan and that she is only wasting her time with him.

Ishaan is not too bothered and blames Anushka for the breakup. He moves into a new rental apartment. His new neighbor Gauri (Prachi Desai) comes into his life. Ishaan begins to fall in love with Gauri as they continue to cross paths and get to know each other. Gauri is confident and playful and like Ishaan she also likes to win at any cost. When Nisha finds out that Ishaan has broken up with Anushka, she comes to Mumbai to stay with him. She later tells Ishaan that she has left his dad (Amar Talwar) as he is driving her bonkers after retirement.

Ishaan finds yet another talent Amala (Sheena Shahabadi) for the music company, one that he believes can be a winner. Ishaan grooms Amala and mentors her in style, fashion and song choices, all the things needed to be a big star in the music industry. Ishaan presents her to Beena. Beena again shoots it down saying that she does not have a promising voice and refuses to sign Amala's contract. While Ishaan is working on his music, Anushka is pregnant with his child but decides not to tell him because he is irresponsible. Anushka had reached out to Shivani, who calls Ishaan for dinner, but he ends up at a pub with Gauri instead. When Shivani tells Ishaan that Anushka is also at dinner, he thinks Anushka wants him back and brings Gauri along with her to make Anushka jealous.

After leaving Shivani's place, where he doesn't speak to Anushka, Ishaan tries to sleep with Gauri, but she pushes back. Ishaan leaves her in the middle of nowhere and goes home. The next day they make up and Gauri explains that she sees Ishaan as a friend.

Ishaan is livid when Beena ignores Amala and launches an old find of Ishaan, Sukanya, as her own find. Ishaan says that he won't support Beena, and she cleverly accepts his resignation. Ishaan is forced out of his company, and Gauri encourages him to start his own music company & launch Amala through that. Ishaan works very hard on the project, but ignores any suggestions that Amala has on her own launch.

But after few days, Ishaan comes to know (Shivani tells Ishaan when she finds that he is getting more serious with Gauri) that Anushka is pregnant with his child. Shivani even escalates to Nisha when Ishaan refuses to accept that it is his child. Shivani finally loses her cool and admonishes Nisha for covering for Ishaan all through his life. Nisha realizes her mistakes and slaps Ishaan.

Gauri gets a scholarship to Paris, and Ishaan and Gauri decide to live there after his big launch. Nisha leaves back for Pune and decides to stop protecting Ishaan anymore. On the day of the launch Ishaan decides to agree with Amala's suggestions. The launch is a massive hit.

Anushka starts having contractions, and Ishaan, realizing his responsibilities, takes Anushka to the hospital where they have a baby girl. Gauri leaves for Paris, understanding what is happening. Eight months later, Ishaan and his baby girl Diya talk to Gauri, who is returning soon, while Anushka is happily married to another guy.
