- Duke was a 4th year surgery student at Parkland Hospital in Dallas on the day that President John F. Kennedy was assasinated. Duke could tell it was futile assisting the doctors working on Kennedy so he turned his attentions to the man across the hall who also had a bullet wound. That man turned out to be Texas governor John Connally.
- He attended Texas A&M University, and received a divinity degree at Southwestern Baptist Theological Seminary. After reading a book about Albert Schweitzer, he reconsidered his career and earned a medical degree at the University of Texas Southwestern Medical Center. He joined the faculty of the 2-year-old University of Texas Medical School at Houston in 1972. In the mid-1970s, he founded the Life Flight air ambulance service in Houston.
- He grew up in Hillsboro, Texas, where he picked cotton, dug ditches, and delivered the Dallas Morning News and the Saturday Evening Post. He enjoyed hunting and fishing in the surrounding countryside.
