The movie is about Desmond. It's about simplifying the narrative. Desmond's brother and father play a significant role in shaping Desmond to being the man he is with the principles he has. He nearly killed his brother when they were children by hitting him in the head with a brick. This caused Desmond to swear to god never to raise a hand in violence again. Later on, when his father was in a drunken rage, Desmond nearly shoots him. This makes him swear to never touch a gun again.
In terms of the film, Desmond's sister may have been away at school or have simply moved away to get away from their father.
